Responding to FDA 483s and Warning Letters Training

Online training course to answer FDA 483 observations and warning letters and bring a site back to compliance.

This practitioner training course starts where inspection readiness ends: at the close of an FDA drug CGMP inspection. It explains what a Form 483 is and is not, how to plan and write a response within 15 business days, how to scope each observation, find root causes and build CAPA plans with evidence, how FDA classifies inspections, and how to answer a warning letter, prepare for regulatory and post-warning letter meetings, and reach a close-out letter. A sterile injectables site in Singapore runs through the course.

About this course

Why the response matters to your company

For a site that supplies the US, the weeks after an FDA inspection can decide its future in that market. FDA has said it generally allows 15 business days for a response to Form 483 observations, and plans a detailed review of a response received in that time before deciding whether to issue a warning letter. A warning letter brings an official action indicated classification, an unacceptable profile and a letter that stays public on fda.gov. For a site outside the US, an import alert can stop shipments: in 2026, two of the cases in this course were placed on the Import Alert 66-40 Red List before their warning letters were dated. FDA's Form 483 FAQ is also clear that a 483 is not a final agency determination: it is weighed with the inspection report, the evidence collected and the firm's response. What the site writes, and what it then does, shapes every one of these outcomes.

What makes this course different

The course starts where inspection readiness ends, at the closing meeting, and runs all the way to the close-out letter. It is built on FDA's own procedures: the Regulatory Procedures Manual, the Investigations Operations Manual 2026, FDA's 2009 notice on post-inspection responses, the June 2025 guidance on post-warning letter meetings under GDUFA, and Import Alert 66-40 as currently published. One sterile site, JKL Pharma in Singapore, is followed from its Form 483 through its response, monthly updates, warning letter, reply, meeting and reinspection to close-out, with every document filled in. Real FDA warning letters from 2025 and 2026 show what responses were found to lack: copies of revised procedures, protocols, interim controls, timelines, and transparency. Each module sets a common mistake against what good looks like, and four practice exercises with model answers let you scope an observation, write a response section, map a warning letter's requests and build a commitment tracker.

Who should take it

The course is for site heads, quality heads, QA and compliance managers, regulatory affairs staff and the owners of investigations and CAPA at drug, API and contract manufacturing sites that supply the US, wherever they are based. It suits teams that have just received a Form 483, sites preparing for an inspection that want to know what follows it, and the executives who sign responses and fund remediation. FDA's manual reminds responsible officials that they have a legal duty to make sure their products and practices comply, so leaders benefit as much as the people who write the documents.

What you can do afterwards

After the course, you can set up a response team on the day the 483 is issued, read each observation for its full scope, and write one complete response, with six headings for every observation, dated actions and indexed exhibits. You can keep FDA informed with updates that report delays before their dates, read a warning letter part by part and answer every request within 15 working days, and judge whether a site is eligible for a post-warning letter meeting. You can also keep a file for every commitment, so that the follow-up inspection can verify each one, and keep the gains after close-out. The scenario-based final exam leads to the Sterling Academy Certificate of Competence for those who score 60 percent or more, with one retake included. It is a training certificate, not an FDA approval.

Who should take FDA 483 response training?

It is for site heads, quality heads, QA and compliance managers, regulatory affairs staff and the owners of investigations and CAPA at drug, API and contract manufacturing sites that supply the US. It suits teams that have just received a Form 483, sites preparing for an FDA inspection, and executives who sign responses and fund remediation.

How long do we have to respond to an FDA Form 483?

FDA says it will generally allow 15 business days to respond to Form 483 observations before deciding on a warning letter, and that it will not ordinarily delay a warning letter to review a response that arrives later. A response is voluntary, but a complete one within that time, with evidence, is what FDA reviews in detail.

How are learners assessed in the FDA 483 response course?

Practice exercises after key modules let you apply each topic to your own organisation and get feedback; they do not count toward the result. The course ends with a scenario-based multiple-choice final exam. Learners who score 60% or more receive a Sterling Academy Certificate of Competence, and one retake is included. Each certificate carries a unique number and a QR code for online checking.

How is this course different from FDA inspection readiness training?

Inspection readiness training prepares a site for the inspection itself. This course starts at the close of the inspection, with the Form 483, and goes in depth into the response, warning letters, meetings, import alerts and close-out. It teaches US FDA requirements only. The certificate is a training certificate for the learner and is not approved by FDA.

When will Responding to FDA 483s and Warning Letters Training be available?

The course is in preparation. Register on this page with “Notify me when it launches” and we will send you one e-mail when it opens for enrolment, with the price per learner and the details for team enrolment. The course page will then show the full module list and the video runtime.
